    A novel crystalline coordination polymer containing Ce3+ and bis(4-nitrophenyl) phosphate (L), CeL3 , was synthesized and its unique transmetalation selectivities toward Yb3+ and Lu3+ in the lanthanide series were evaluated. The relatively large difference in transmetalation selectivity between the neighboring Tm3+ and Yb3+ species is noteworthy because the reactivities of heavy lanthanides are generally considerably similar. The structural strain of the polymeric framework is likely responsible for this unusual trend. Powder X-ray diffraction analysis indicated that, in the cases of only Yb3+ and Lu3+ , large differences in their ionic sizes compared to that of Ce3+ in the parent framework may induce a structural strain after solid solution formation, while cleavage of the relatively weak Ce-O bond allows the formation of new Yb-O and Lu-O bonds with the incoming Yb3+ and Lu3+ , respectively. Structural phase transitions likely caused by the heterogeneous nucleation of the Yb- (or Lu-) type phase were also observed.

    放射性廃棄物の処分に向けて、廃棄物試料中のアクチニド(Th、U、Np、Pu、Am、Cm)を簡易かつ高感度に分析する方法を開発することを目的として、キャピラリー電気泳動-レーザー励起蛍光検出法(CE-LIF)の適用性について検討している。本検討では、ウランを分析対象として、ウラニルイオンと安定な錯体を形成する配位骨格を有する蛍光プローブを新規に合成し、CE-LIFによりウラニルイオンの分離検出を試みた。その結果、溶液試料において、従来のCE法の検出限界(数十ppb-ppmレベル)を大幅に改善した数十pptレベルの検出限界を達成できた。

    Analytical methods of Ln^III were developed in which the kinetic inertness of Ln^III complexes with acyclic polyaminocarboxylate (PAC) on the dissociation reaction was the key to establishing detection systems. First, a pre-derivatization HPLC method with fluorescent detection was developed for closedshell Ln^III using an aromatic octadentate PAC as the derivatizing reagent. Selective detection at ppb levels was achieved. Second, the factors to form inert Ln-pac complexes were investigated on the dissociation reaction. The rate constants and activation parameters were determined for a series of Ln^III-hexadentate and octadentate PAC complexes. Interestingly, an unusually large negative activation entropy for solvolysis governed the kinetic inertness, and was probably derived from metal envelopment in the complex and hydration on the activation state. Third, a novel measurement method of the dissociation rate was developed using capillary electrophoresis (CE). The dissociation rate constant for the Ce^III-PAC complex was determined for the first time. Fourth, based on the result of the second study, a sensitive pre-derivatizing CE method for Ln^III at the fmol level was developed using an aromatic EDTA derivative. In this method, a high resolution was achieved by introducing a ternary complex equilibrium in the carrier. Finally, the energy-transfer luminescence of a Tb^III complex with a PAC possessing phenol function was studied. The detection limit at the ppt level was achieved, and the photophysics was also investigated.

    This study aims to investigate the function of the gas phase and the supercritical fluid incorporated in porous hydrophobic packing materials in the columns of surface bubble-modulated liquid chromatography and supercritical fluid stationary phase liquid chromatography and develop unprecedented separation technologies. The effects of the structure of the interfacial liquid layer formed on the surface of the hydrophobic packing materials and the chemical structure of the surface of the hydrophobic materials on the separation selectivity were clarified. We also carried out the research with the aim of developing a method for determining volatile organic compounds in an aqueous solution and a high-speed analysis method for inert gases that does not require pretreatment by gas-liquid separation.

    We developed a simple, rapid, and low-cost simultaneous on-site indicator bacteria measurement technique using fluorescent probes. A sample with 40 cfu/L of E. coli could be detected in about 7 hours, and a sample with 4 cfu/L in about 13 hours. The operation is extremely simple: even with untreated sewage, the sample water is placed in a liquid medium or agar medium without any pretreatment, and the fluorescence intensity is automatically measured once every 10 minutes using a microplate reader set at 37°C. Since the number of coliform bacteria can be measured in about three hours for normal treated sewage water, the number of coliform bacteria can be analyzed with high frequency in both time and space, and deterioration of treatment efficiency can be quickly identified. Since the measurement medium is about 0.02 mL, the cost per sample is about 2 Japanese yen.

    In this research project, the micro-particle-assisted capillary electrophoresis (MACE) system discovered by the applicant was introduced into a nucleic acid aptamer selection experiment (SELEX) system for various target molecules to demonstrate that there is still a large room for growth in the SELEX system, which has been attempted to be improved using a synthetic organic chemistry approach. The objective of this study was to demonstrate that the SELEX method can be significantly improved by using an "analytical chemistry" approach. As a result, we demonstrated that the MACE-SELEX method works on a wide range of targets, including soluble proteins, membrane proteins, and small molecules, and that it is possible to obtain DNA aptamers with high binding affinity.

    Highly sensitive and rapid analytical method for ultratrace actinide ions in radioactive samples by capillary electrophoresis-laser-induced fluorescence detection (CE-LIF) was developed. We designed emissive probes for the detection of actinide ions with ppt levels using CE-LIF. This method was successfully applied to real radioactive liquid samples collected from nuclear facilities, including the Fukushima Daiichi Nuclear Power Plant.

    We developed surface-bubble-modulated liquid chromatography (SBMLC), which has a hybrid stationary phase consisting of gas phase as well as the hydrophobic material and the water/hydrophobe interface and showed that this chromatographic method can be performed with the columns packed with various types of hydrophobic porous materials. We determined the distribution coefficients of various organic compounds referring to accumulations onto the water/alkyl chain interface and into the alkyl chain layer from the bulk water by SBMLC. The experimental data provide a picture of the spatial distribution of organic molecules in alkyl bonded silica particles exposed to water. We also developed a liquid chromatography with supercritical fluid stationary phase, which has the hybrid stationary phase consisting of the supercritical carbon dioxide phase, the interfacial liquid phase and the hydrophobic moiety.

    A new chromatographic method termed surface-bubble-modulated liquid chromatography, that has a hybrid separation medium incorporated with surface nanobubbles, was developed. Nanobubbles can be fixed at the interface between water and a hydrophobic material by delivering water into a dry column packed with a nanoporous material. The incorporation of a gas phase leads to the formation of the hybrid separation system consisting of the gas phase, hydrophobic moieties, and the water/hydrophobic interface or the interfacial water. One can change the volume of the gas phase by pressure applied to the column, which in turn alters the area of water/hydrophobic interface, while the amount of the hydrophobic moiety remains constant. Therefore this strategy provides a novel technique not only for manipulating the separation selectivity by pressure but also for elucidating the mechanism of accumulation or retention of solute compounds in aqueous solutions by a hydrophobic material.

    A polyacrylamide gel electrophoresis (PAGE)-based method has been developed, consisting of two gel electrophoresis, to obtain distribution of protein-bound metal ions in biological samples. The former is blue-native (BN) PAGE to separate proteins without dissociation during migration. The other is a PAGE for the separation and detection of protein-bound metal ions in small volume samples with high sensitivity in the ppt range using fluorescence metal probe. The probes have a chelating moiety (multidentate polyaminocarboxylate), a fluorophore (fluorescein) and a spacer (aminobenzyl group) to form emissive and kinetic inert complexes with metal ions. Some metal ions, Mg^<2+>, Cu^<2+>, Fe^<2+>/^<3+>, Ni^<2+>, Co^<2+>, Mn^<2+>, Cd^<2+> and Hg^<2+>, were successfully detected by this new PAGE. After separation of the proteins by BN-PAGE, the metal ions in the gel fractions were eluted, followed by derivatization of copper ions into the metal probe complexes to be separated and determined by fluorescence detection in the second PAGE. In this PAGE-based method, the copper ions bound to ceruloplasmin and superoxidedismutase, and iron ions bound to transferrin and hemopexin were quantitatively determined.

    Novel fluorescent probes for detecting metal ions working in capillary electrophoresi-laser-induced fluorescence detection (CE-LIF) and polyacrylamide gel electrophoresis (PAGE) have been developed. The detection limits for several transition metal ions of tens pM were successfully obtained. In CE-LIF, the addition of cationic polymer provided high resolution among metal-probe complexes with new separation principle. The new separation mode was named as "Ion association complex separation mode." In PAGE, Fe and Cu ions which existed in the gel fractions of transferrin and ceruloplasmin were successfully determined.

    Ion-exchange chromatography with superheated water, i.e., liquid water under pressure at temperatures above 100 °C (SW-IEC) was developed and the mechanism of ion-exchange separation processes was investigated using SW-IEC. Based on the results obtained, it was demonstrated that SW-IEC is a powerful tool for changing separation selectivity for ionic solutes drastically by controlling temperature. The states and separation function of the interfacial water on hydrophobic surfaces were clarified and an HPLC method was presented for determination of equilibrium constants and kinetic constants of chemical reactions proceeding on the hydrophobic surfaces as well as in the bulk solution phases. Furthermore redox derivatization two-dimensional HPLC system was developed using catalytic redox activity of porous graphitic carbon and it was shown that this separation technique is very useful for selective separation and determination of trace amounts of reducible and oxidizable compounds.

    A microchip capillary electrophoretic reactor (MCER), a practical and powerful tool for dissociation kinetic analysis of metal complexes, has been proposed. In the dissociation kinetic analysis of metal complexes using conventional batch reaction techniques, owing to the difficulty in dividing between reactant (metal complex) and products (metal ion and free ligand), the direct measurement of the dissociation kinetics had not been achieved. On the contrary, MCER revolutionized the measurement of the dissociation rate constant of metal complexes since its accomplishment of the direct measurement. The basic idea of a MCER was provoked by the concentration jump character of the sharp band profiles in microchip capillary electrophoresis (MCE) ; i.e. metal complexes during the MCE separation processes, in which there had been no ligand in the electrophoretic buffer solution, were exposed to overwhelming force to dissociation caused by a steep concentration jump of the ligand. The time depending dissociation reaction profile of metal complex of Ce^<3+> with a polyaminocarboxylic ligand, 8-amino-2-[(2-amino-5-methylphenoxyl)methyl]-6-methoxyquinoline-N,N,N',N',-tetraacetic acid ([CeL]^-) was described using both internal and external standard. The solvolytic dissociation rate constant of [CeL]^-have been determined directly as 6.2 X 10^<-3> s^<-1> at 303 K with MCER.
    Capillary electrophoretic reactor (CER) was applied to the determination of dissociation reaction rate constant of bio-molecular complexes. Both an immunocomplex of anti digoxin IgG with digoxin and a complex double stranded DNA with ethidium bromide were chosen as analytical model compounds. Unfortunately, determination of dissociation reaction rate constants could not be achieved with CER in both systems, since their properties of small electrophoretic mobility and lower kinetically inertness.
